This CLIA kit uses the Sandwich-CLIA principle. The micro CLIA plate provided in this kit has been pre-coated with an antibody specific to Mouse GT-34. Samples or Standards are added to the micro CLIA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse GT-34 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse GT-34, biotinylated detection antibody and Avidin-HRP conjugate will appear luminescence. The Relative light unit (RLU) value is measured by the Chemiluminescence immunoassay analyzer at 425nm. The RLU value is positively associated with the concentration of Mouse GT-34. You can calculate the concentration of Mouse GT-34 in the samples by comparing the RLU value of the samples to the standard curve.
This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Mouse GT. During the reaction, Mouse GT in the sample or standard competes with a fixed amount of Mouse GT on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Mouse GT. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Mouse GT in tested samples can be calculated by comparing the OD of the samples to the standard curve.
